Collagen

Collagen is the most abundant protein in our body, about 30% of total protein. Collagen is rich in bone, skin, tendon, ligament, cartilage, heart, eye, and so on. Synthesis of collagen protein is one of the most complicated processes among all human proteins. Many enzymatically posttranslational modifications involve the synthesis such as 3- and 4-hydroxyproline and 5-hydroxylysine. Some of hydroxylysine residues are additionally modified as galactosyl-hydroxylysine and glucosyl-galactosyl-hydroxylysine. 4-Hydroxylation of proline increases the thermal stability of triple helical structure. These posttranslational modification reactions are essential to the biological function. Defection of these enzymes causes abnormality in bone and other organs. Due to the many posttranslational modifications and the large size of molecule, it is impossible to produce collagen molecules from recombinant expression with reasonable prices. With Nippi’s more than a half century of research expertise, we produce purified collagen from various kinds of animal tissues such as skin, and cornea. NMP Collagen PS has antigenic telopeptides removed by pepsin extraction and subsequent advanced purification and is widely used as a medical raw material. It is also highly soluble directly in water only, as the powder has been adjusted to a slightly acidic state. Only raw porcine hides that have been issued a “Slaughterhouse Inspection Certificate “by Japan’s Meat Hygiene Inspection Office are used as raw material, thus proving its safety under strict control. The manufacturing process includes virus inactivation processes to ensure the safety of the products.

Types I, III, and V collagens are available. For type I collagen, both acid soluble and pepsin solubilized are available from skin. Acid soluble collagen has noncollagenous telopeptides at both the N- and C- termini, and reconstitutes fibrils that more closely resemble native physiological collagen fibrils. Type III collagen is abundant in dermis and aorta. In skin, about 80% of fibrillar collagen is type I, and the 15-20% is type III collagen. We have the experience, knowledge, and skill regarding collagen protein extraction. Custom orders from different animal species and different types of collagen are also available upon request. This product is a reconstituted collagen fiber sheet. This sheet is made from highly purified collagen extracted from porcine dermis with pepsin. The sheet is cross-linked and has high strength. This is sterilized with ethylene oxide gas and is ready for cell culture. Various cells can be cultured on the sheet, and the high strength is maintained even after culturing, so please use it as a base material for cell transplantation.
